Abstract Translated from German Verfahren zur Isolierung und Reinigung von Amino-, glycosid-Antibiotika der Formel <IMAGE> insbesondere zur Reinigung von Sisomicin, 5-Episisomycin, Metilmicin, Gentamycin und 3"-N-Desmethyl-3"-N-Ethylsisomicin, durch ein mehrstufiges Aufarbeitungsverfahren, das sich einer vorteilhaften Konzeption zur Isolierung und Reinigung von Aminoglykosiden bedient: Sie kombiniert eine selektive Lipophilisierung der betreffenden Verbindung der Formel I im fermentativ gewonnenen Rohprodukt mit gezielter Flüssig-Flüssig-Extraktion dieses lipophilisierten Produktes. Die Lipophilisierung wird dadurch erreicht, daß in den Verbindungen der Formel I enthaltene Aminogruppen mit geeigneten Schutzgruppen versehen werden. Nachdem das gewünschte Aminoglykosidderivat durch Flüssig-Flüssig-Extraktion in reiner Form abgetrennt worden ist, werden die Schutzgruppen wieder abgespalten, und man erhält in das gewünschte Aminoglykosid der Formel I in reiner Form. Process for the isolation and purification of amino, glycoside antibiotics of the formula <IMAGE>, in particular for the purification of sisomicin, 5-episisomycin, metilmicin, gentamycin and 3 "-N-desmethyl-3" -N-ethylsisomicin, by a multi-stage work-up process, which uses an advantageous concept for the isolation and purification of aminoglycosides: it combines a selective lipophilization of the compound of formula I in question in the crude product obtained by fermentation with targeted liquid-liquid extraction of this lipophilized product. Lipophilization is achieved in that amino groups contained in the compounds of the formula I are provided with suitable protective groups. After the desired aminoglycoside derivative has been separated off in pure form by liquid-liquid extraction, the protective groups are split off again, and the desired aminoglycoside of the formula I is obtained in pure form.
