Abstract
Abstract Recombinant microbial cloning vehicles comprising heterologous DNA coding for the expression of mammalian hormones (e.g., somatostatin) and other polypeptides, in- duding plasmids suited for the transformation of bacterial hosts are prepared. The latter incorporate a regulon homologous to the host in its untransformed state, in reading phase with the structural gene for the heterologous DNA.

Claims Analysis
Total Claims: 10
Claim 1
Independent Claim
74 words
A recombinant plasmid suited for transformation of a hactexial host and use therein as a cloning vehicle, characterised in that the plasmid comprises: a) A regulon homologous to the bacterial host in its untransformed state; and by In reading phase with the regulon, a DNA insert coding for the amino acid sequence of a heterologous polypeptide, such that bacteria transformed by the plasmid are capable of expressing said amino acid sequence in recoverable form.
Claim 2
Dependent Claim
7 words
A bacterial plasmid according to claim 1.
Claim 3
Dependent Claim
22 words
A plasmid according to claim 2 whose regulon is essentially identical to a regulon ordinarily present in the bacterial host's chromosomal DNA.
Claim 4
Dependent Claim
33 words
A plasmid according to claim 2 wherein the regulon comprises the promoter - operator system of an operon selected from the group consisting of E. coli. lac operon and E. coli. tryptophan operon.
Claim 5
Dependent Claim
22 words
A plasmid according to any one of the preceding claims wherein the heterologous polypeptide is a mammalian hormone or an intermediate therefor.
Claim 6
Dependent Claim
38 words
A plasmid according to claim 5 wherein the heterologous polypeptide is human preproinsulin, human proinsulin, the A chain of human insulin, the B chain of human insulin, human or bovine growth hormone, leutinizing hormone, ACTH or pancreatic polypeptide.
Claim 7
Dependent Claim
14 words
A plasmid according to claim 5 wherein the heterologous polypeptide is a mammalian hormone.
Claim 8
Dependent Claim
12 words
A plasmid according to claim 5 wherein the heterologous polypeptide is somatostatin.
Claim 9
Dependent Claim
21 words
A plasmid according to claim 8 wherein the regulon comprises the promoter - operator system of the E. coli. lac operon.
Claim 10
Dependent Claim
29 words
A plasmid according to any one of the preceding claims for use in the transformation of a bacterial host to render said host capable of expressing a heterologous polypeptide.
