Abstract

Abstract The present invention involves an improved test system for the determination of glucose in aqueous fluids of the type which comprises glucose oxidase, a peroxidatively active substance and chromogen. The improvement involves the use of m-anisidine as a chromogen which permits the semi-quantitative determination of glucose at higher concentrations than is possible with previously used chromogens.

Claims Analysis

Total Claims: 7
Claim 1
Independent Claim 87 words
In a test composition capable of the determination of the presence of glucose in a liquid test sample, wherein said composition comprises glucose oxidase, a peroxidatively active substance and one or more chromogens reactive with hydrogen peroxide, and wherein the components of the composition are present together as a solution and are present in amounts and proportions such that the composition is capable of differentiating between concentrations of 2,000 and 5,000 milligrams of glucose per deciliter, the improvement which involves the use of m-anisidine as a chromogen.
Claim 2
Dependent Claim 87 words
In a test composition capable of the determination of the presence of glucose in a liquid test sample, wherein said composition comprises glucose oxidase, a peroxidatively active substance and one or more chromogens reactive with hydrogen peroxide, and wherein the components of the composition are present together as a solution and are present in amounts and proportions such that the composition is capable of differentiating between concentrations of 2,000 and 5,000 milligrams of glucose per deciliter, the improvement which involves the use of m-anisidine as a chromogen.
Claim 3
Dependent Claim 47 words
The test composition of claim 1 which is in the form of a liquid reagent solution containing 1,000 to 1,600 international units of glucose oxidase per milliliter, 600 to 1,300 international units of horseradish peroxidase per milliliter and from 10 to 15 milligrams per milliliter of m-anisidine.
Claim 4
Dependent Claim 53 words
The test composition of claim 1 which contains a second chromogen having a high extinction coefficient in an amount sufficient to provide a system capable of detecting the presence of 100 milligrams of glucose per deciliter while retaining the ability to differentiate between concentrations of 2,000 and 5,000 milligrams of glucose per deciliter.
Claim 5
Dependent Claim 23 words
The test composition of claim 3 wherein the second chromogen is present in an amount of from 0.5 to 1.0 milligram per milliliter.
Claim 6
Dependent Claim 17 words
The test composition of claim 3 wherein the second chromogen is benzidine, syringaldazine, diaminofluorine, o-tolidine or tetramethylbenzidine.
Claim 7
Dependent Claim 31 words
The test composition of claim 1 which contains a buffer in sufficient quantity to maintain the pH of the liquid test sample at a level of from about 5.0 to 7.0.
