Abstract

Abstract The histamine released by mast cells on contact with an allergen can be determined by measuring the uptake by the mast cells of a fluorescent acridone, which uptake is proportional to the amount of histamine released by action of the antigen. Fluorescence of the acridone absorbed by the mast cells is measured in conventional manner and the amount of histamine released is determined thereby.

Claims Analysis

Total Claims: 9
Claim 1
Independent Claim 63 words
A METHOD OF DETERMINING THE SUSCEPTIBILITY OF A SUBJECT TO ALLERGENS WHICH COMPRISES THE STEPS OF INCUBATING MAST CELLS WITH SERUM OF THE SUBJECT AT TEMPRATURES OF 30* TO 45*C., CONTACTING THE INCUBATED MAST CELLS WITH AN ALLERGEN AND THEREATER ADDING A DILUTE SOLUTION OF AN ABSORBABLE FLUORESCENT ACRIDONE TO THE ALLERGEN TREATED CELLS AND MEASURING THE ACRIDONE ABSORBED BY THE MAST CELLS.
Claim 2
Claim 3
Dependent Claim 30 words
The method of claim 1 in which 100,000 to 1,000,000 mast cells are incubated for 1 to 5 minutes with 0.05 to 0.5 ml. of blood serum of a subject.
Claim 4
Dependent Claim 34 words
The method of claim 2 in which the mast cells are suspended in isotonic saline at a pH within the range 6.8 to 7.2 while in contact with the blood serum, allergen and acridone.
Claim 5
Dependent Claim 42 words
The method of claim 1 in which the acridone absorbed by the mast cells is recovered therefrom by lysis and the amount of the acridone absorbed by the mast cells and recovered therefrom is determined by observing the fluorescence of the acridone.
Claim 6
Independent Claim 81 words
A method of measuring the amount of histamine released from activated mast cells in response to their reaction to an allergen while suspended in isotonic saline at pH 6.8 to 7.2 which comprises the steps of allowing the mast cells to absorb a fluorescent acridone from a dilute aqueous isotonic solution thereof, recovering the mast cells from the liquid in which they are suspended, lysing the mast cells to release the fluorescent acridone and measuring the fluorescence of said absorbed acridone.
Claim 7
Independent Claim 81 words
A method of measuring the amount of histamine released from activated mast cells in response to their reaction to an allergen while suspended in isotonic saline at pH 6.8 to 7.2 which comprises the steps of allowing the mast cells to absorb a fluorescent acridone from a dilute aqueous isotonic solution thereof, recovering the mast cells from the liquid in which they are suspended, lysing the mast cells to release the fluorescent acridone and measuring the fluorescence of said absorbed acridone.
Claim 8
Dependent Claim 14 words
The method of claim 5 in which the fluorescent acridone is 2, 10-dimethylaminopropyl acridone.
Claim 9
Dependent Claim 12 words
The method of claim 5 in which the fluorescent acridone is acridone.
