Abstract
Abstract According to the method of this invention, highly pure teneicine can be obtained in a high yield from an extract of an extracellular substance containing teneicine by flowing said extract through an affinity chromatograph containing gelatin bound thereto via covalent bonds to remove fibronectin to such an extent that its content is lowered to 1 % by weight or less of the solute and purifying the fraction thus treated by using a basic anion exchange resin.

Claims Analysis
Total Claims: 10
Claim 1
Independent Claim
77 words
A process for purifying tenascin characterized in that the process comprises the steps of removing fibronectin from extracellular stromal extracted materials containing tenascin so that the content ratio of the fibronectin is not more than 1 % by weight on the basis of the total solute of the effluent by using affinity chromatography media containing gelatin covalently attached to a matrix and then applying a run-through fraction on a basic anion exchange resin to purify the tenascin.
Claim 2
Dependent Claim
32 words
A process according to claim 1, wherein the extracellular stromal extracted materials are derived from a cell selected from the group consisting of human-fibroblast cells, human-melanoma cells, human-glyoma cells, and human-chondrosarcoma cells.
Claim 3
Dependent Claim
30 words
A process according to claim 1, wherein the extracellular stromal extracted materials are eluted with an extracting agent selected from the group consisting of urea, guanidine hydrochloride and guanidine isotiocyanate.
Claim 4
Dependent Claim
19 words
A process according to claim 2, wherein the extracellular stromal extracted materials are obtained from a cell culture medium.
Claim 5
Dependent Claim
19 words
A process according to claim 1, wherein the basic anion exchage resin is a resin modified with diethylaminoethyl groups.
Claim 6
Dependent Claim
30 words
A process according to claim 1, wherein the purification by the basic anion exchange resin is affected by using not more than 1 M sodium chloride in 1-8 M urea.
Claim 7
Dependent Claim
30 words
A process according to claim 1, wherein the purification by the basic anion exchange resin is affected by using not more than 0.4 M sodium chloride in 2 M urea.
Claim 8
Dependent Claim
30 words
A process according to claim 1, wherein the purification by the basic anion exchange resin is affected by using a linear ascending gradient of 0-0.4M sodium chloride in 2M urea.
Claim 9
Dependent Claim
23 words
A process according to claim 8, wherein the purification by the basic anion exchange resin is affected by a high performance liquid chlomatography.
Claim 10
Dependent Claim
16 words
A process according to claim 1, wherein tenascin is detected by tenascin dependent cell attachment activity.
