Process for producing proteins and polypeptides
Abstract
The invention relates to a process for the production of proteins by cocionation of DNA sequences which encode a biospecific protein or polypeptide (I) and a further protein (II) which encode the protein to be produced or a marker protein, these two DNA sequences via one Linked DNA sequence, which encodes a short-chain peptide (III), which acts as a specific cleavage site and this peptide contains one or more amino acids in addition to the cleavage sequence, which increase the cleavage specificity and / or cleavage rate. This new piece of DNA is introduced into a host organism by means of suitable genetic engineering methods so that transcription, translation and expression can take place. The combined protein thus obtained is purified by affinity chromatography and then cleaved enzymatically.
