Abstract

Abstract Improved dyes are provided comprising phycobiliproteins having covalently coupled thereto a plurality of small molecular weight organic dyes under conditions permitting energy transfer mechanisms to operate therebetween. The resultant label may then be used in virtually any immunoassay format.

Claims Analysis

Total Claims: 12
Claim 1
Independent Claim 38 words
1 . A fluroescent label comprising at least one organic dye molecule coupled to a phycobiliprotein whereby excitation of said organic dye molecule results in energy transfer to said phycobiliprotein, resulting in detectable fluorescent emission by said phycobiliprotein.
Claim 2
Dependent Claim 38 words
1 . A fluroescent label comprising at least one organic dye molecule coupled to a phycobiliprotein whereby excitation of said organic dye molecule results in energy transfer to said phycobiliprotein, resulting in detectable fluorescent emission by said phycobiliprotein.
Claim 3
Dependent Claim 19 words
The label of Claim 1, wherein there are a plurality of organic dye molecules covalently coupled to said phycobiliprotein.
Claim 4
Dependent Claim 25 words
The label of Claim 1 or Claim 2, wherein the spectral emission characteristic or said organic dye molecule overlaps the excitation spectrum of said phycobiliprotein.
Claim 5
Dependent Claim 24 words
The label of any one of claims 1 to 3, wherein the organic dye molecule has a molecular weight from 100 to 1000 daltons.
Claim 6
Dependent Claim 18 words
The label of any one of Claims 1 to 4, wherein the organic dye molecule is Azure A.
Claim 7
Dependent Claim 15 words
The label of any one of Claims 1 to 5, wherein the phycobiliprotein is allophycocyanin.
Claim 8
Dependent Claim 27 words
A fluorescent label comprising a plurality of Azure A molecules covalently coupled to allophycocyanin, said coupling permitting energy transfer between said Azure A molecules and said allophycocyanin.
Claim 9
Dependent Claim 27 words
A fluorescent label comprising a plurality of Azure A molecules covalently coupled to allophycocyanin, said coupling permitting energy transfer between said Azure A molecules and said allophycocyanin.
Claim 10
Dependent Claim 24 words
A ligand-ligand binding receptor pair wherein one member of the pair is associated with the label of any one of claims 1 to 7.
Claim 11
Dependent Claim 57 words
The use of the label of any one of claims 1 to 7 or the ligand-ligand binding receptor pair of claim 8 in an assay for the detection of a ligand by detecting an immunological reaction between said ligand and a ligand binding receptor by detecting said label associated with the ligand or the ligand binding receptor.
Claim 12
Dependent Claim 32 words
The use of Claim 9, wherein the label is illuminated at a first frequency capable or exciting said organic dye and fluorescent emission from said phycobiliprotein is detected at a second frequency.
