Abstract

Abstract A system is disclosed for treating blood, with the objective of reducing the functioning population of a blood constituent. According to the method, blood is passed through an ultraviolet radiation field in the presence of a dissolved photoactive agent capable of forming photo- adducts with receptors on the constituent to effect covalent bonding between the photoactive agent and the same. If desired, the blood to be treated may be separated to provide a blood fraction rich in the constituent sought to be reacted and low in any blood component which absorbs UV radiation, as for example by continuous centrifuging, which fraction is then irradiated with UV radiation in the presence of the dissolved photoactive chemical agent.

Claims Analysis

Total Claims: 10
Claim 1
Independent Claim 83 words
A system for treating blood in order to reduce the functioning population of a blood constituent by associating and reacting said constituent with a photoactive chemical agent characterized by: (a) means for receiving said blood and subjecting said blood to a density gradient whereby red blood cells are substantially separated from said blood constituent; and (b) a source of UV radiation oriented so as to preferentially impinge radiation upon said blood constituent so as to react the associated chemical agent with said constituent.
Claim 2
Independent Claim 83 words
A system for treating blood in order to reduce the functioning population of a blood constituent by associating and reacting said constituent with a photoactive chemical agent characterized by: (a) means for receiving said blood and subjecting said blood to a density gradient whereby red blood cells are substantially separated from said blood constituent; and (b) a source of UV radiation oriented so as to preferentially impinge radiation upon said blood constituent so as to react the associated chemical agent with said constituent.
Claim 3
Dependent Claim 63 words
The system of claim 1 wherein said means comprise a centrifuge which functions to separate said blood into a blood fraction comprising a substantial portion of said constituent and having a concentration of said red blood cells less than that of untreated blood and wherein said system further comprises means for transporting said fraction into a blood treatment chamber comprising said radiation source.
Claim 4
Dependent Claim 36 words
The system of claim 1 wherein said means comprise a centrifuge having the source of UV radiation internally and substantially centrally located therein whereby said blood may be simultaneously subjected to said density gradient and irradiated.
Claim 5
Dependent Claim 66 words
The system of claim 1 wherein said photoactive chemical agent comprises an agent specific for receptor sites in or on the blood constituent and capable when activated by said UV radiation of forming photo-adducts with blood constituent receptor sites, to thereby effect chemical bonding between said photoactivated chemical agent and said receptor sites, thereby initiating the destruction and/or removal of said blood constituent from the blood.
Claim 6
Dependent Claim 39 words
The system according to claim 4 wherein the photoactivated chemical agent comprises a polypeptide selected from the group consisting of insulin, interleukin, thymopoietin and transferrin, and wherein said agent further comprises a photocytotoxic chemical agent covalently-linked to said polypeptide.
Claim 7
Dependent Claim 49 words
The system according to claim 4 wherein the photoactivated chemical agent comprises a liposome, said liposome having an antibody or a polypeptide capable of binding to said constituent receptor site covalently linked to its outer surface, and said liposome internally incorporating an effective amount of a photocytotoxic agent therein.
Claim 8
Independent Claim 44 words
The system according to claims 5 or 6 wherein the photocytotoxic chemical agent comprises a moiety selected from the group consisting of psoralen, pyrene cholesteryloleate, acridine, prophyrin, fluorescein, rhodamine, 16-diazocortisone, ethidium, transition metal complexes of bleomycin, transition metal complexes of deglycobleomycin and organoplatinum complexes.
Claim 9
Independent Claim 166 words
A system for treating blood with a photoactive chemical agent which comprises: (a) a source of UV radiation; (b) a blood treatment chamber adapted to receive radiation from said source whereby said photoactive chemical agent may be activated; (c) and continuous transport means adapted to transport blood from a reservoir through said blood treatment chamber and back to said reservoir, wherein said photoactive chemical agent is specific for a receptor site in or on the blood constituent and capable when activated by said UV radiation of forming photoadducts with said receptor site, to thereby effect chemical bonding between said photoactivated chemical agent and said receptor site, thereby initiating the destructlon and/or removal of said blood constituent from the blood supply of the subject and wherein said chemical agent is selected from the group consisting of (a) a polypeptide covalently linked to a photocytotoxic chemical and (b) a liposome, said liposome having a polypeptide or antibody covalently bound thereto and said liposome incorporating a photocytotoxic chemical therein.
